dc.description.abstractRezumat Hepatotoxicitatea preparatelor antituberculoase constituie un obstacol major în realizarea unui tratament eficient și inofensiv al tuberculozei, determinat de riscul afectării separate a fi catului, care crește mult la asocierea acestora. Studiul aspectelor patogenetice ale hepatotoxicității preparatelor antituberculoase a demonstrat că aceasta este cauzată de: formarea metaboliților intermediari toxici; stimularea activității enzimelor hepatice responsabile de generarea produselor toxice; declanșarea stresului oxidativ, cu diminuarea activității sistemului antioxidant; inducerea proceselor inflamatoare și imune; stimularea apoptozei și a necrozei hepatocitelor; prezența devierilor genetice, responsabile de metabolismul preparatelor antituberculoase; dereglarea funcțiilor organitelor intracelulare etc. Elucidarea verigilor patogenetice ale hepatotoxicității preparatelor antituberculoase va permite de a selecta rațional și a administra adecvat medicamentele hepatoprotectoare pentru prevenirea și/sau tratamentul afecțiunilor hepatice.en_US
dc.description.abstractHepatotoxicity of antituberculosis drugs (literature synthesis) Hepatotoxicity of antituberculosis drugs is an important obstacle to effective and harmless treatment of tuberculosis, determined by the risk of separate liver damage, which greatly increases when they are associated. The study of the pathogenetic aspects of the hepatotoxicity of antituberculosis drugs has shown that this is caused by: formation of toxic intermediates metabolites; stimulation of the activity of the liver enzymes responsible for generating toxic products; triggering oxidative stress by decreasing activity of the antioxidant system; induction of inflammatory and immune processes; stimulation of hepatocyte apoptosis and necrosis; the presence of genetic deviations, responsible for the metabolism of antituberculosis preparations; disruption of the functions of intracellular organisms, etc. The elucidation of the pathogenetic links of the hepatotoxicity of the antituberculosis preparations will allow to rationally select and target the hepatoprotective drugs for its prevention and / or treatment.
dc.description.abstractГепатотоксичность противотуберкулезных препа- ратов (обзор литературы) Гепатотоксичность противотуберкулезных препа- ратов является важным препятствием для эффек- тивного и безопасного лечения туберкулеза, обуслов- ленного риском как самостоятельным поражением печени каждым лекарством, так и при их сочетании. Изучение патогенетических аспектов гепатотоксич- ности противотуберкулезных препаратов показало, что это обусловлено: образованием промежуточных токсических метаболитов; стимуляцией активности ферментов печени, ответственных за образование токсичных продуктов; развитием окислительного стресса и снижением активности антиоксидантной системы; индукцией воспалительных и иммунных про- цессов; стимуляцией апоптоза и некроза гепатоцитов; наличием генетических нарушений, ответственных за метаболизм противотуберкулезных препаратов; нарушением функций внутриклеточных органел и др. Выяснение патогенетических звеньев гепатотоксич- ности противотуберкулезных препаратов позволит рационально выбрать и целенаправленно назначить гепатопротекторы для профилактики и/или лечения поражений печени.
