There was elaborated the TLC method of alprazolam identification by using of five&#13;
develop systems, spectrophotometric method of quantitative determination for apprazolam in&#13;
powder and tablets of 0.001 g were elaborated, as well as HPLC method was elaborated in order&#13;
to determine the purity of substance.Using the analytical methods elaborated by us over the&#13;
artificial biological test there was extracted alprazolam from biological liquids (blood plasma,&#13;
urine). Having considered the elaborated methods as specific and reproductive they a&#13;
rerecommended for the study of alprazolam in biological liquids.&#13;

A fost elaborată metoda cromatografiei pe strat subţire de identificare a alprazolamului,&#13;
utilizând 2 sisteme de solvenţi, metoda spectrofotometrică de dozare în pulbere şi comprimate&#13;
0.001 g, precum şi metoda HPLC de identificare în pulbere. Aplicând metodele analitice&#13;
elaborate de noi la probele biologice „in vitro” s-a efectuat extracţia alprazolamului din lichidele&#13;
biologice. Dat fiind faptul, că metodele folosite sunt specifice şi reproductive ele se recomandă&#13;
pentru studierea alprazolamului în lichidle biologice.

Optimization of the HPLC method for analysis of Milk-Thistle fruit (Silybum marianum (L.)&#13;
Gaertn).&#13;
An interlaboratory irreproducibility of the results of Milk-Thistle fruit assay has been&#13;
discovered at realization of analytical procedure by the European Pharmacopoeia. It was found,&#13;
that this irreproducibility is caused by insufficient plenitude of the analytes extraction from&#13;
vegetal material.&#13;
A new procedure of probe preparation, that consists of two sequential extractions by ethylic&#13;
alcohol 40% for 60 min at 70°С, has been developed. The offered procedure ensures recovery&#13;
about 98 % and essentially decreases the volume of work.&#13;
Use of acetonitrile in the mobile phase composition and optimization of the gradient shape&#13;
or switch-over to isocratic elution are offered for decrease of the analysis time.&#13;
Key words: Silybum marianum, flavonolignans, HPLC-UV, pharmaceutical analysis.&#13;

La efectuarea analizei cantitative a fructelor de armurariu prin metoda Farmacopeei&#13;
Europene s-a depistat o ireprodictibilitate interlaborator al rezultatelor. S-a stabilit că aceasta&#13;
ireproductibilitate este condiţionată de insuficienţa plenitudinii de extracţie a analiţilor din&#13;
produsul vegetal.&#13;
S-a elaborat un nou procedeu de preparare a probelor, care constă din două extracţii&#13;
consecutive cu alcool etilic 40% timp de 60 min la temperatura 70°C. Procedeul propus asigură&#13;
regăsirea circa 98% şi micşorarea esenţială a volumului de lucru.&#13;
Pentru reducerea timpului analizei cromatografice s-a propus utilizarea acetonitrilului în&#13;
componenţa fazei mobile şi optimizarea formei gradientului sau trecerea la eluare izocratică.

The purpose of this research was to develop mouth dissolve tablets of Chlorpheniramine&#13;
maleate. Tablets were prepared by direct compression. The tablets were evaluated for&#13;
disintegration time and hardness test. In the investigation, a 32&#13;
 full factorial design was used to&#13;
investigate the joint influence of 2 formulation variables: amount of Croscarmellose sodium and&#13;
Lactose. The results of multiple linear regression analysis revealed that for obtaining of a rapidly&#13;
disintegration dosage form, tablets should be prepared using an optimum concentration of&#13;
Lactose and higher percentage of Croscarmellose sodium.&#13;

Au fost cercetate comprimatele orodispersabile cu maleat de clorfeniramină, preparate prin&#13;
comprimare directă. A fost evaluat timpul de dezagregare şi rezistenţa mecanică. Pentru&#13;
optimizarea formulării a fost folosit designul factorial deplin randomizat 32&#13;
. Croscarmeloza&#13;
sodică şi lactoza au fost selectate ca variabile independente.&#13;
În rezultatul analizei multiple regresionale a fost relevat raportul optimal al cantităţilor de&#13;
croscarmeloză şi lactoză care permit obţinerea valorilor scontate ale timpului de dezagregare şi&#13;
al rezistenţei mecanice a comprimatelor cu dezagregare rapidă.

There was elaborated optimum isolation and extraction conditions of amitriptiline from&#13;
biological fluids. We used physico – chemical methods to identify amitriptiline: chromatography&#13;
on a thin layer by utilizing four systems of solvents, spectrophotometrical method in UV,&#13;
HPLC.The methods processed by us for in biological tests “in vitro” are specific and&#13;
reproductive, and they are recommended for the study of amitriptiline in biological fluids.&#13;

Au fost determinate condiţiile optime de izolare şi extragere a amitriptilinei din lichidele&#13;
biologice Am aplicat metodele fizico-chimice de identificare a amitriptilinei: cromatografia pe&#13;
strat subţire utilizând patru sisteme de solvenţi, metoda spectrofotometrică în UV, HPLC.&#13;
Metodele prelucrate de noi pentru amitriptilină în probele biologice „in vitro” sunt specifice şi&#13;
reproductive şi ele se recomandă pentru studierea amitriptilinei în lichidele biologice.
