Introducere. Taraxacum officinale F. H. Wigg (TO) reprezintă
o sursă importantă de compuși biologic activi cu diverse
funcţii, inclusiv antioxidantă. Reactivitatea înaltă a grupelor
tiolice implică aceștia în calitate de primii antioxidanţi consumaţi
în cazul stresului oxidativ. Scopul acestui studiu a fost de
a evalua mecanismele de acţiune ale diferitor extracte din TO
asupra conţinutului de tioli eritrocitari.
Material și metode. În studiu au fost utilizate rădăcini
(TOR) și frunze uscate de TO (TOF) extrase pe etanol de diverse concentraţii (10%, 20%, 25%, 40%, 50% și 80%) și dimetil
sulfoxid (DMSO). Conţinutul de tioli a fost evaluat în conformitate
cu metoda Erel et al. (2014), în modificaţia noastră.
Rezultate. În cazul extractelor etanolice din TOR, conţinutul
grupelor SH-libere (μM/g Hb) a fost evaluat după cum
urmează: în alcool de 10% – 3,21±0,11, 20% – 3,58±0,08,
25% – 2,88±0,04, 40% – 2,91±0,06, 50% – 3,18±0,07 și în
80% – 3,08±0,03. Aceeași grupă de tioli în cazul extractului
cu DMSO a fost cuantificată ca 3,23±0,23. Conţinutul grupelor
SH-totale a fost diferit în funcţie de concentraţia etanolului:
10% – 3,36±0,13, 20% – 3,72±0,11, 25% – 3,19±0,12, 40%
– 3,01±0,01, 50% – 3,38±0,17 și în 80% – 3,24±0,07. În extractul
din TOR pe DMSO nivelul tiolilor totali a fost evaluat cu
3,53±0,18.
Conţinutul grupelor SH-libere s-a modificat sub influenţa
extractelor din TOF, precum: în etanol de 10% – 3,20±0,23,
20% – 3,24±0,03, 25% – 3,04±0,16, 40% – 2,88±0,06, 50%
– 3,05±0,09, 80% – 3,02±0,01, iar în DMSO – 3,30±0,31.
Cantitatea grupelor SH-totale: în extractele pe alcool de
10% – 3,39±0,25, 20% – 3,47±0,05, 25% – 3,28±0,04, 40%
– 3,12±0,02, 50% – 3,26±0,12, 80% – 3,15±0,02 și 3,79±0,08
sub influenţa TOF pe DMSO. Analiza comparativă a pus în evidenţă
doar două extracte etanolice din TOF (20% și 25%),
care au crescut conţinutul de SH. TOF s-a dovedit a fi capabil
de a menţine concentraţia ridicata de tioli eritrocitari, fapt neobservat
în cazul TOR.
Concluzii. Studiile noastre anterioare și actuale demonstrează,
că frunzele și rădăcinile din Taraxacum officinale sunt
o sursă valoroasă de diverse substanţe biologic active, implicate
în diferite mecanisme biochimice, inclusiv protecţia antioxidantă.
Extractele din TO influenţează asupra nivelului tiolic
eritrocitar, care este dependent de tipul extractantului și concentraţia
acestuia. În general, influenţa TO ţine de micșorarea
nivelului de tioli, mecanism care depinde probabil de conţinutul
formelor redusă și oxidată din probele experimentale,
precum și de concentraţiile, combinaţiile substanţelor biologic
active în diferite tipuri de extracte. Cu toate acestea mecanismul
activităţii sale antioxidante rămâne neclar și necesită
studii suplimentare.
Introduction. Taraxacum officinale F. H. Wigg (TO) represents
an important source of biologically active compounds
with different functions, including antioxidative. The high reactivity
of thiol groups involves them as first antioxidants consumed
in case of oxidative stress. The aim of this research was
to evaluate the mechanisms of action of different TO extracts
on RBC’s thiols content.
Material and methods. TO dried roots (TOR) and leaves
(TOL) extracts on ethanol (10%, 20%, 25%, 40%, 50% and
80%) and dimethyl sulfoxide (DMSO) were used. The thiols nivecontent
was evaluated in accordance with Erel et al. (2014)
method, modified by us.
Results. In case of TOR ethanol extracts, native-SH content
(μM/g Hb) was as follows: 10% – 3,21±0,11, 20% – 3,58±0,08,
25% – 2,88±0,04, 40% – 2,91±0,06, 50% – 3,18±0,07 and
in 80% – 3,08±0,03. The same thiols were evaluated as
3,23±0,23μM/g Hb in TOR DMSO extracts. The total-SH
amount was also different. In ethanolic extracts it changed as
follows: in alcohol of 10% – 3,36±0,13, 20% – 3,72±0,11, 25%
– 3,19±0,12, 40% – 3,01±0,01, 50% – 3,38±0,17 and in 80%
– 3,24±0,07. In case of roots extracted with DMSO total thiols
level was evaluated as 3,53±0,18. The content of native-SH (μM/g Hb) changed under the TOL
influence, extracted with ethanol of different concentration, as
follows: 10% –3,20±0,23, 20% – 3,24±0,03, 25% – 3,04±0,16,
40% – 2,88±0,06, 50% – 3,05±0,09 and in 80% – 3,02±0,01.
The same thiols were evaluated as 3,30±0,31 under the DMSO
leaves extracts influence. The total-SH amount changed under
the influence of ethanol extracts as follows: in alcohol of
10% – 3,39±0,25, 20% – 3,47±0,05, 25% – 3,28±0,04, 40% –
3,12±0,02, 50% – 3,26±0,12 and 3,15±0,02 in ethanol extracts
of 80%. In case of DMSO leaves extracts total thiols level was
evaluated as 3,79±0,08. Overall analysis revealed only two TOL
ethanolic extracts of (20% and 25%), that increased SH-level,
compared to all other extracts. TOL abound in antioxidants
compounds capable of maintaining the high concentration of
thiols in erythrocytes, but the roots extracts do not possess this
capacity.
Conclusions. Our previous and present studies demonstrate
that Taraxacum officinale leaves and roots are a valuable
source of different classes of biologically active substances,
involved in various biochemical mechanisms, including antioxidant
protection. TO extracts influence on RBC’s thiols level
depends of type of extractant and its concentration. The overall
influence of TO is to decrease the thiols level, mechanism
which probably depends on their reduced and oxidized forms
level in experimental samples, as well on various concentrations
and combinations of biologically active substances in different
types of extracts. However, the mechanism of its antioxidant
activity remains unclear and requires further studies.
