Introducere. Micetele din genul Candida se regăsesc ca floră normală la indivizii sănătoși și sunt implicate în etiologia infecțiilor oportuniste cu rate mari de mortalitate, în special la persoanele imunocompromise. Speciile acestui gen sunt micetele cel mai frecvent implicate în patologia umană datorită factorilor de virulență ce îi posedă și capacității de dezvoltare a rezistenței la antifungice. Scopul lucrării. Elucidarea rezistenței la antifungice și a factorilor enzimatici de patogenitate a tulpinilor clinice de Candida albicans. Material și metode. Au fost examinate 87 tulpini de C. albicans izolate din diverse biosubstrate clinice, care au fost identificate prin tehnici microbiologice standarde. Testarea sensibilității la antifungice s-a efectuat conform metodologiei EUCAST (The European Committee on Antimicrobial Susceptibility Testing) prin metode cantitative cu determinarea concentrației minime inhibitorii (E-test, Vitek 2 Compact). Determinarea enzimelor de patogenitate s-a efectuat conform metodologiei în vigoare. Rezultate. Tulpinile de C. albicans studiate au prezentat niveluri diferite de sensibilitate la antimicoticele testate. Cel mai înalt nivel de rezistență a fost înregistrat la miconazol (52,2%) și itraconazol (34,6%). Tulpinile de C. albicans au prezentat sensibilitate la chetoconazol (91,8%) și flucitosină (89,6%). La studierea enzimelor de patogenitate s-a constatat că 65,2% dintre tulpini au manifestat activitate lipolitică și 73,5% - activitate hemolitică. Concluzii. Rezultatele studiului denotă o rată înaltă de rezistență la unele preparate antifungice a tulpinilor clinice de C. albicans. Enzimele proteolitice și hemolizinele, factori importanți de patogenitate responsabili de invazia și distrugerea țesuturilor gazdei, au fost puși în evidență la peste 60% dintre tulpini.
Background. Candida is found as normal flora in healthy individuals and are involved in the etiology of opportunistic infections with high mortality rates, especially in immunocompromised individuals. Species of this genus are the fungi that are most frequently involved in human pathology due to their virulence factors and their ability to develop antifungal resistance. Objective of the study. Elucidation of antifungal resistance and enzymatic factors of pathogenicity of clinical strains of Candida albicans. Material and Methods. 87 C. albicans strains isolated from various clinical biosubstrates were examined and identified by standard microbiological techniques. Antifungal susceptibility testing was performed according to EUCAST methodology (The European Committee on Antimicrobial Susceptibility Testing) by quantitative methods with the determination of the minimum inhibitory concentration (E-test, Vitek 2 Compact). The determination of enzymes’ pathogenicity was performed according to up-to-date methodology. Results. The C. albicans strains studied showed different levels of sensitivity to the tested antifungals. The highest levels of resistance were to miconazole (52.2%) and itraconazole (34.6%). C. albicans strains are susceptible to ketoconazole (91.8%) and flucitosin (89.6%). When studying the pathogenic enzymes, it was found that 65.2% of the strains showed lipolytic activity and 73.5% - hemolytic activity. Conclusion. The results of the study show a high rate of resistance to some antifungal drugs of clinical strains of C. albicans. Proteolytic enzymes and hemolysins, important pathogenic factors responsible for the invasion and destruction of host tissues have been identified in over 60% of strains.
